Technical data sheet (Babesia bovis)
Technical details for
ready to use PCR kit for Babesia bovis (gel agarose); 100 Reactions
Bovine babesiosis is a tick-borne disease found worldwide in cattle, which is
caused by Babesia. After the acute infection (primary infection), recovered
animal carries this blood parasite subclinically and it makes these animals
undetectable with microscopic diagnosis. These animals serve as carriers,
which act later as reservoir for the infection in the herd and continue
infecting the tick vectors, which spread the parasite. Therefore it is
essential to screen the animals (carriers) with highly sensitive and specific
tests. Microscopic diagnosis can not offer such sensitivity as well as
serological tests have the problem of cross reactions. The cell culture needs
too long time for detection and needs cell culture laboratory. Therefore PCR
is merging as gold standard in the field of medicine as the PCR tests are
highly sensitive, specific as well as can detect Babesia bovis in one drop of
blood also. The PCR can be also used for animals, which are infected and
showing symptoms for differential diagnosis.
target gene: highly conserved region of RNA gene sequence of Babesia bovis
Kind of samples: blood samples (blood samples can be shipped throughout the
world as one can put 2-3 drops of blood on filter paper as dried blood is
sufficient to run the test and send in a letter to any laboratory in the world:
It is advisable to put this in extra plastic bag), Tissue and other body
fluids. This can be used to screen the bovine serum to be used in cell culture
preparation, which can be later on a part of vaccine or monoclonal antibodies
production. This needs isolated DNA, which can be isolated with commercial
available kit or home made method.
Validation: It has been checked for the cross reactions with related and non
related micro organisms like Anaplasma, Neospora caninum, Babesia bigemina,
Toxoplasma gondii, DNA of healthy catte, Borrelia burgdorferi, Mycobacterium
bovis, Leptospira, Brucella abortus, Theileria etc. The serial dilution has
been done to see the detection limit. The specificity of the test is 96-98%.
Detection limit: upto one CPU unit in reaction

negative control, loading dye, gene marker (100 bp), PCR master mix and manual.
