Helicobacter pylori (HP) is a gram-negative spirally shaped bacterium and is
playing a role during the pathogenesis of active chronic gastritis and peptic
ulcer disease of humans. It has been also detected in animals like dogs etc.
To eradicate this bacterium, one has to identify this in the samples and there
are a number of methods available to detect this bacterium like serological
methods. The serological methods have the problem of cross reactions as well
as low sensitivity. However one can use other methods like Staining and
culture methods. The culture method is time consuming as it grows slowly,
therefore the method use should be PCR method, where one can hope to detect
the bacterium in the smallest samples with highest sensitivity and specificity.
Target gene: highly conserved region of HP
Samples: gastric biopsy, tissue, ulcer, other body fluid samples, bacterial
cultures, rectal swabs (in dogs) etc. This assay needs the isolated good
quality DNA, which can be isolated with good commercially available kits as
well as home made method.
Key point: This assay has been checked for the cross reactions with related
and non related micro organisms like Helicobacter mustelae, Campylobacter
jejuni, Ureaplasma, Mycoplasma genitalium etc.
Components: primers, polymerase, nucleotides, buffer, molecular water, loading
dye, molecular marker (gene ruler), negative control, positive control and
manual. In order to use the internal control: one can use the Genekam species
specific DNA identification kit for human for human probes or canine for dog
probes.
