Legionella spp. (gel agarose) for 100 reactions
Legionella spp. are gram-negative bacteria responsible to cause respiratory
disorders like pneumonia after the inhalation of water droplets. There are
around 42 species, which comprise 64 serogroups of legionellae and most of
them may cause the disease. About 3-7% community acquired pneumonia is caused
through Legionella spp. Particularly elderly and immunocomprised patients are
at great risk of death. Clinical presentation of legionellosis involves kidney,
liver, gastrointestinal and central nervous system dysfunctions. To eradicate
this bacterium, one has to identify this in the samples and there are a number
of methods available to detect this bacterium like serological methods. The
serological methods have the problem of cross reactions as well as low
sensitivity. However one can use other methods like staining and cell culture
methods. The culture method is time consuming and needs special medium
therefore the method use should be PCR method, where one can hope to detect
the bacterium in the smallest samples with highest sensitivity and specificity.
One can get the results on the same day.
Target gene: highly conserved region of RNA genes of Legionella: This gene
exists as multiple copies per genome.
Samples: blood, serum, plasma, respiratory swabs, tissue, other body fluid
samples, cell cultures, water probes, drainage probes etc.. This assay needs
the isolated good quality DNA, which can be isolated with good commercially
available kits as well as home made method.
Key point: This assay has been checked for the cross reactions with related
and non related micro organisms like Staphylococcus, Candida albicans,
Haemophilus influenzae, Bordetella, Chlamydia pneumoniae, Mycoplasma
pneumoniae etc.
This assay can detect clinical relevant Legionella spp. like L. pneumophila,
L. micdalei, L. longbeachae, L. bozemanii, L. feacalis and L. dumoffii etc.
Components: primers, polymerase, nucleotides, buffer, molecular water, loading
dye, molecular marker (gene ruler), negative control, positive control and
manual. In order to use the internal control: one can use the Genekam species
specific DNA identification kit for human for human probes or other species.
Detection limit: around 2 CFU (10 fg) per reaction
