Mycobacterium avium sp. paratuberculosis (MAP) is known to cause the chronic
disease called Johne´s disease in domestic as well as wild ruminants. The
symptoms of the disease are chronic diarrhoae along with decreased milk
production, aneamia, infertility and even death in final stage of disease. The
infected animals are shedding heavy number of bacteria in the feaces and
contaminating the enviroment, therefore it is essential to remove the diseased
or carrier animals. It is also shed in milk. MAP is known to play a role in
human beings suffering from immunosuppressive diseases like HIV as the
presence of MAP can complicate the case of such patients. Mycobacteria are
Gram positive and acid-fast bacteria. Because of subclinical infections, this
pathogen has been undiagnosed for serval years. There is a discussion going
about the role of MAP in Crohn´s disease in human beings, hence making this
bacterium as an important food safety corcern. There are also reports that MAP
can survive some form of pasteurization, therefore there is a need of
affective diagnostic as well as control measurements of this bacterium.
Therefore it is essestial to detect this bacterium as early as possible. In
order to detect this, there are different methods like staining, culture
growth as well as serolgical methods available, but these methods have
problems like low sensitivity, needs very long time as well as cross reactions
etc, therefore PCR method is merging as the gold standard as it is highly
sensitive, highly specific and results can be obtained in 24 hours. In case of
MAP, many laboratories are targetting the genesequence IS900 of MAP to detect
this as this IS900 has been found in other mycobacteria species, hence it
leads to cross reactions and lower specificity. Even there are reports of
identication of enviromental mycobacteria with IS900 PCR, therefore Genekam
Biotechnology AG decided to bring the PCR kit on the market to target 2
different genes in order to give the most accurate detections for MAP as PCR
with only IS900 may give false results or results of IS900 PCR should be taken
with caution. Even this problem can not be solved with restriction
endonucleasis analysis. Hence the ready to use PCR (double check) for MAP is
the answer.

Samples: feaces, blood, milk, tissue, digestive tract, lymphnodes and other
body parts. This kit needs isolated good DNA, which can done through
commercial available kit or home made method.
Validation: It has been checked for cross reactions with related or non
related micro organisms: different Mycobacteria, Borrelia burgdorferi,
Leptospira, Coxiella burnetii, Bovine Herpes virus and many more. It has been
checked for serial dilutions. It has been checked to detect the MAP strains
from different cattles.
