
Technical details for ready to use PCR kit for Mycoplasma hyorhinis (gel agarose based; 100 reactions)
Mycoplasma hyorhinis is a pathogenic
mycoplasma, which causes respiratory disorders (called enzootic pig pneumonia:
a chronic respiratory disease) in pigs world wide along with M. hyopneumoniae.
This is characherized by high morbidity rates as well as low mortality leading
to economic losses through retarded growth, poor food conversion and increased
susceptibility of pigs to other infections, therefore there is a need of
laboratory diagnosis. This laboratory diagnosis can be done through culture
methods (which takes a longtime as Mycoplasma goes slowly and needs special
media), serological methods like ELISA as well as FAT, which are prone to
cross reactions, therefore the PCR is merging as a goldstandard for detection
of micro organisms in medicine as this is highly sensitive and specific also.
Moreover PCR needs a little material to conduct the test. Moreover PCR can be
used to differentiate between the different strains of M. hoypneumoniae, M.
hyorhinis and M. flocculare.
Target gene: highly conserved region of gene of Mycoplasma hyorhinis.
Kinds of samples: respiratory system swabs, nasal swabs, colonies grown on
cultures, blood (serum/plasma) samples, tissue samples and other body fluid
samples. This kit needs isolated DNA, which can be isolated using commercial
available kits or home made methods.
Components of the kit: polymerase, forward and backward primers, nucleotides,
buffers, PCR master mix, loading dye, gene ruler (100 bp), positive control,
negative control as well as manual.
Validations: It has been tested for cross reactions with related and non
related micror organisms like Mycoplasma hyopneuomiae, Chlamydia psittaci,
healthy pig DNA, Acholeplasma laidlawii and many more. It is tested for serial
dilutions.
Sensitivity: 1-50 pg of extracted Mycoplasma hyorhinis DNA
