Technical details for ready to use PCR kit for Mycoplasma meleagridis (gel agarose based; 100 reactions)

Mycoplasma meleagridis is a pathogenic mycoplasma causing heavy mortality turkey, there is a transmission of this bacteria through eggs also. This causes heavy economical losses along with Mycoplasma iowae as they cause sinusitis, airsinusitis along with the bone abnormal development. Moreover the mycoplasmas can be present in the enviromental samples also. They cause disease in poultry birds also, therefore there is a need of laboratory diagnosis. This laboratory diagnosis can be done through culture methods (which takes a longtime as Mycoplasma goes slowly and needs special media), serological methods like ELISA as well as FAT, which are prone to cross reactions, therefore the PCR is merging as a goldstandard for detection of micro organisms in medicine as this is highly sensitive and specific also. Moreover PCR needs a little material to conduct the test. PCR can be used to detect them in even one day old chickens.

Target gene: highly conserved region of gene of Mycoplasma meleagridis

Kinds of samples: eggs, respiratory swabs, enviromental probes, water probes, colonies grown on cultures, blood (serum/plasma) samples, tissue samples and other body fluid samples. This kit needs isolated DNA, which can be isolated using commercial available kits or home made methods.

Components of the kit: polymerase, forward and backward primers, nucleotides, buffers, PCR master mix, loading dye, gene ruler (100 bp), positive control, negative control as well as manual.

Validations: It has been tested for cross reactions with related and non related micro organisms like Mycoplasma synoviae, M. iowae, M. gallisepticum, M. gallinarum, M. pullorum, Chlaymdia psittaci, NDV, IBR, different Staphylocci, Staphylococcus aereus, different Streptococci, E.coli and many more . It is tested for serial dilutions.
